Bike won't start after carb cleaning

So when I bought my bike, the previous owner had been running it on the choke. I figured a jet was clogged or maybe air was leaking somewhere so I pulled the carbs, cleaned them and put everything back. I put in some gas, primed it for 20 seconds or so and all it would do was crank. For some reason I decided to set it to prime afterwards and try to start it like that, now it won't even crank. If it does it's only for a brief second.

So, obviously I'm going to have to get it to crank again first. Can anybody tell me what went wrong by trying to start the engine on prime?
